A green, biomimetic, phosphate-mediated Pictet-Spengler reaction was used in the synthesis of three catecholic tetrahydroisoquinolines, 1, 2, and 12, present in the medicinal plant Portulaca oleracea, as well as their analogues 3-11, 13, and 14, with dopamine hydrochloride and aldehydes as the substrates. AB-8 macroporous resin column chromatography was applied for purification of the products from the one-step high efficacy synthesis. It eliminated the difficulties in the isolation of catecholic tetrahydroisoquinolines from the aqueous reaction system and unreacted dopamine hydrochloride. Activity screening in CHO-K1/G alpha 15 cell models consistently expressing alpha(1B)-, beta(1)-, or beta(2)-adrenergic receptors indicated that 12 and 2, compounds that are present in P. oleracea, possessed the most potent beta(2)-adrenergic receptor agonist activity and 2 was a selective beta(2)-adrenergic receptor agonist at the concentration of 100 mu M. Both 12 and 2 exhibited dose-dependent bronchodilator effects on the histamine-induced contraction of isolated guinea-pig tracheal smooth muscle, with EC50 values of 0.8 and 2.8 mu M, respectively. These findings explain the scientific rationale of P. oleracea use as an antiasthmatic herb in folk medicine and provide the basis for the discovery of novel antiasthma drugs.

The first example of an asymmetric Guerbet reaction has been developed. Using commercially available, classic Noyori Ru-II-diamine-diphosphine catalysts, well-known in asymmetric hydrogenation, racemic secondary alcohols are shown to couple with primary alcohols in the presence of a base, affording new chiral alcohols with enantiomeric ratios of up to 99:1. Requiring no reducing agents, the protocol provides an easy, alternative route for the synthesis of chiral alcohols. Mechanistic studies reveal that the reaction proceeds via a Ru-catalyzed asymmetric hydrogen autotransfer process in concert with a base-promoted allylic alcohol isomerization.

Monoclonal antibodies (mAb) are a major focus of the pharmaceutical industry, and polyclonal immunoglobulin G (IgG) therapy is used to treat a wide variety of health conditions. As some individuals require mAb/IgG therapy their entire life, there is currently a great desire to formulate antibodies for bolus injection rather than infusion. However, to achieve the required doses, very concentrated antibody solutions may be required. Unfortunately, mAb/IgG self-assembly at high concentration can produce an unacceptably high viscosity for injection. To address this challenge, this study expands the concept of dynamic covalent chemistry to dynamic bioconjugation in order to reduce viscosity by interfering with antibody-antibody interactions. Ultra-sub-stoichiometric amounts of dynamic PEGylation agents (down to the nanomolar) significantly reduced the viscosity of concentrated antibody solutions by interfering with oligomerization.

Residual chemicals in wooden children’s products may threaten children’s health, but few studies have paid attention to it so far. In this study, a simple and green method was developed for the determination of volatile chemicals, especially the 48 fragrance allergens in wooden children’s products using static headspace coupled with gas chromatography and mass spectrometry. Minimal amounts of sample (20 mg) and acetone solvent (40 mu L) were used, and no additional sample pretreatment was performed except for freezing and grinding. Three factors (incubating temperature, equilibration time, pressing pressure) affecting headspace extraction were optimized by response surface methodology to obtain scientific results considering of the interactions among variables. Under the optimum detection conditions, this method was successfully validated for the analysis of fragrances. The proposed method showed limits of quantification within the range of 0.2-20 mg/kg. Recoveries ranged from 79.8 to 115.2%, and the relative standard deviation (n = 5) varied from 0.6 to 9.9%. Moreover, interferences caused by different sample matrices were investigated. The method was further applied to the analysis of 30 commercial wooden samples. Results demonstrated the existence of fragrances in wooden children’s products. Benzyl alcohol, trans-2-heptenal, eugenol, and isoeugenol were the compounds with high detection rate.

Hepatocellular carcinoma (HCC) is one of the most prevalent cancers worldwide. Nowadays, pharmacological therapy for HCC is in urgent needs. Paclitaxel is an effective drug against diverse solid tumors, but commonly resisted in HCC patients. We recently have disclosed that microtubule affinity-regulating kinase 4 (MARK4) increases the microtubule dynamics and confers paclitaxel resistance in HCC, suggesting MARK4 as an attractive target to overcome paclitaxel resistance. Herein, we synthesized and identified coumarin derivatives 50 as a novel MARK4 inhibitor. Biological evaluation indicated compound 50 directly interacted with MARK4 and inhibited its activity in vitro, suppressed cell viability and induced apoptosis of HCC cells in a MARK4-dependent manner. Importantly, compound 50 significantly increased the drug response of paclitaxel treatment to HCC cells, providing a promise strategy to HCC treatment and broadening the application of paclitaxel in cancer therapy.

An ultrasonic-assisted synthesis of bis-isoxazole derivatives was developed. Eight 3-(6-chloropyridin-3-yl)-5-{[(3-arylisoxazol-5-yl)methoxy]methyl}isoxazoles were synthesized by 1,3-dipolar cycloaddition reaction between substituted isoxazolyl alkyne compounds and 6-chloro-N-hydroxynicotinimidoyl chloride. The structures of the synthesized compounds were confirmed by HRMS, FTIR, H-1 and C-13 NMR spectroscopy. Wherein, the antifungal and antibacterial activities of target compounds were tested. The synthesized compounds 6a and 6h exhibited better antifungal activity in comparison with the standard drug itraconazole. The minimum inhibitory concentrations(MICs) of both compound 6a and compound 6h were both 4 mu g/mL against Candida albicans ATCC 10231.

Under metal- and ligand-free conditions, the economical and accessible CF3SO3H successfully catalyzed homogeneous O-H bond insertion reactions between hydroxyl compounds and diazo compounds. Including phenols, alcohols, water and oximes, these O-H bond insertion reactions are very general and functional-group tolerant. Here, we first report the O-H bond insertion of oximes (ketoximes and aldoximes), and their structures were characterized by X-ray crystallography. Moreover, a simple and effective method for S-H insertion reactions of thiols was also developed, delivering the desired C-S bond products with good to excellent yields. It is worth noting that the efficacy of the developed methodology could be further shown by the expeditious synthesis of the PPAR agonist MBX-102 acid.

In this study, we present a versatile and easy procedure for modifying a cobalt ferrite nanoparticle step by step. A new nanocatalyst was prepared via Cu(II)immobilized onto CoFe2O4@HT@Imine. The catalyst was fully characterized by Fourier-transform infrared (FT-IR), energy-dispersive X-ray spectroscopy (EDX), field emission scanning electron microscopy (FE-SEM), X-ray diffraction (XRD), and vibrating sample magnetometer (VSM) analyses. The current procedure as a green protocol offers benefits including a simple operational method, an excellent yield of products, mild reaction conditions, minimum chemical wastes, and short reaction times. Without any significant reduction in the catalytic performance, up to five recyclability cycles of the catalyst were obtained. The optimization results suggest that the best condition in the oxidation of benzyl alcohol derivatives is 0.003 g of the CoFe2O4@HT@Imine-Cu(II)catalyst, TEMPO, at 70 degrees C under solvent-free condition and air.

In recent years, magnetically active CuFe2O4 nanoparticles have been gaining significant interest in the field of heterogeneous catalysis as those can be easily prepared and effortlessly recovered from a reaction system. Here, we are reporting our work on cascade syntheses of 1,3-dimethyl-6-nitro-5-arylpyrido[2,3-d]pyrimidine-2,4(1H,3H)-diones starting from 6-[(dimethylamino)methylene-amino]-1,3-dimethyluracil, aromatic aldehydes and nitromethane using magnetically active CuFe2O4 catalyst system under microwave irradiation and solvent-less condition. The current methodology is a valued addition to the existing procedures of 5-arylpyrido[2,3-d]pyrimidines syntheses making best use of the diene behavior of 6-[(dimethylamino)methylene-amino]-1,3-dimethyluracil. However, heterogeneous catalysis has been employed for the first time to do the syntheses by carrying out [4 + 2]cycloaddition reaction between 6-[(dimethylamino)methylene-amino]-1,3-dimethyluracil and in situ generated 2-(2-nitrovinyl)arenes/heteroarenes. The methodology is highly time-economic, and along with other features like easy recovery and good reusability of the catalyst, simple operating procedure, wide substrate scope, and good to excellent product yield, it offers the chemists a reaction protocol worth trying for the syntheses of 1,3-dimethyl-6-nitro-5-arylpyrido[2,3-d]pyrimidine-2,4(1H,3H)-diones. While laboratory prepared catalyst system was characterized using FT-IR, XRD, SEM-EDX, VSM, XPS and TEM analysis, all the synthesized compounds have been characterized using H-1 and C-13 NMR spectroscopy and HRMS.

A composite film with multiple anti-counterfeiting features was demonstrated by superposing quantum dots (QDs) polymer matrix (film A) and cholesteric liquid crystal film (film B) together. The first-line and second-line anti-counterfeiting characteristics were successfully implemented by employing thermochromic, angular photochromic, and circularly polarized discoloration of film B, respectively. By initiatively utilizing the different relative positions between the fluorescence emission peak (lambda(em)) of film A and the central selective reflection wavelength (lambda(em)) of film B at different temperatures, which resulted in changes in the fluorescence spectra or the different presence of latent patterns, the most important third-line anti-counterfeiting feature was successfully achieved.
