Identification of Phenoxyalkylbenzimidazoles with Antitubercular Activity

We conducted an evaluation of the phenoxyalkylbenzimidazole series based on the exemplar 2-ethyl-1-(3-phenoxypropyl)-1H-benzo[d]imidazole for its antitubercular activity. Four segments of the molecule were examined systematically to define a structure-activity relationship with respect to biological activity. Compounds had submicromolar activity against Mycobacterium tuberculosis; the most potent compound had a minimum inhibitory concentration (MIC) of 52 nM and was not cytotoxic against eukaryotic cells (selectivity index = 523). Compounds were selective for M. tuberculosis over other bacterial species, including the closely related Mycobacterium smegmatis. Compounds had a bacteriostatic effect against aerobically grown, replicating M. tuberculosis, but were bactericidal against nonreplicating bacteria. Representative compounds had moderate to high permeability in MDCK cells, but were rapidly metabolized in rodents and human liver microsomes, suggesting the possibility of rapid in vivo hepatic clearance mediated by oxidative metabolism. These results indicate that the readily synthesized phenoxyalkylbenzimidazoles are a promising class of potent and selective antitubercular agents, if the metabolic liability can be solved.

PdNPs@P2VP-Fe3O4 organic-inorganic hybrid microgels as a nanoreactor for selective aerobic oxidation of alcohols

The preparation of organic-inorganic hybrid microgels with functional Fe3O4 nanoparticles as the crosslinker and 2-vinyl pyridine (2VP) as the pH-sensitive monomer is reported. Magnetic Fe 3O4 nanoparticles were first synthesized via a redox reaction and then modified with 3-(trimethoxysilyl)propylmethacrylate (TMSPMA) via the silanization. The bonding of multiple TMSPMA monomers on the surface of Fe3O4 nanoparticles renders them as crosslinker. Radical polymerization of 2VP was then carried out with the presence of TMSPMA-modified Fe3O4 nanoparticles, leading to the formation of P2VPFe3O4 hybrid microgels crosslinked with Fe 3O4 nanoparticles. The synthesized hybrid microgels are able to trap metal ions such as Pd2? via complex formation of P2VP with metal ions. The reduction of trapped palladium ions by sodium borohydride led to immobilized-palladium nanoparticles. The synthesized PdNPs@P2VP-Fe 3O4 is employed as a new nanocatalyst toward oxidation reaction of alcohols. In addition, the catalytic activity of metal nanocomposite has been found that can be modulated by the volume transition of microgel structures by pH change for the oxidation reaction of benzyl alcohol. Transmission electron microscopy, thermogravimetric analysis, vibrating sample magnetometer, scanning electron microscopy, X-ray powder diffraction, Atomic absorption and Fourier transform Infrared were then used to characterize the resultant catalyst. Indole-Substituted Benzothiazoles and Benzoxazoles as Selective and Reversible MAO-B Inhibitors for Treatment of Parkinson’s Disease

To develop novel, selective, and reversible MAO-B inhibitors for safer treatment of Parkinson’s disease, benzothiazole and benzoxazole derivatives with indole moiety were designed and synthesized. Most of the synthesized compounds showed inhibitory activities against MAO-B and selectivity over MAO-A. The most active compound was compound 5b, 6-fluoro-2-(1-methyl-1H-indol-5-yl)benzo[d]thiazole with an IC50 value of 28 nM with no apparent effect on MAO-A activity at 10 muM. Based on the reversibility assay, compound 5b turned out to be fully reversible with over 95% of recovery of enzyme activity after washout of the compound. Compound 5b showed a reasonable stability in human liver microsomes and did not affect the activities of CYP isozymes, suggesting an absence of high-risk drug-drug interaction. In an in vivo MPTP-induced animal model of Parkinson’s disease, oral administration of compound 5b showed neuroprotection of nigrostriatal dopaminergic neurons as revealed by tyrosine hydroxylase staining and prevention of MPTP-induced parkinsonism as revealed by motor behavioral assay of vertical grid test. In summary, the novel, reversible, and selective MAO-B inhibitor compound 5b was synthesized and characterized. We propose compound 5b as an effective therapeutic compound for relieving parkinsonism.

Nanostructured RuO2 on MWCNTs: Efficient catalyst for transfer hydrogenation of carbonyl compounds and aerial oxidation of alcohols

Multiwall carbon nanotubes (MWCNTs)/ruthenium dioxide nanoparticles (RuO2NPs) composite was prepared by a straightforward ‘dry synthesis’ method. After being well characterized, the prepared composite was used as a nanocatalyst (RuO2/MWCNT) for the transfer hydrogenation of carbonyl compounds. The excellent adhesion of RuO2NPs on the anchoring sites of MWCNTs was confirmed by TEM and Raman analyses. The weight percentage (7.97 wt%) and the chemical state (+4) of Ru in RuO2/MWCNT was confirmed by EDS and XPS analyses, respectively. It was found that the RuO2/MWCNT has a higher specific surface area of 189.3 m2 g-1. Initially the reaction conditions were optimized and then the scope of the catalytic system was extended with a wide range of carbonyl compounds. The influence of the size of RuO2NPs on the transfer hydrogenation of carbonyl compounds was also studied. The RuO2/MWCNT is highly chemoselective, heterogeneous in nature, reusable and highly stable. Owing to the high stability of the used catalyst (u-RuO2/MWCNT), it was further calcinated at high temperature to obtain RuO2 nanorods (NRs) hybrid MWCNTs. Then the hybrid material was used as a catalyst (r-RuO 2/MWCNT) for the aerial oxidation of alcohols and the result was found to be good.
