Aim: Tumor cells adapt to hypoxic microenvironments by releasing the key transcription factor HIF-1 alpha, which promotes angiogenesis, glycolytic phenotype, metastasis and erythropoiesis, allowing proliferation amid low oxygen levels. Therefore, therapeutic targeting of HIF-1 alpha represents a viable strategy for cancer therapy. Methods & Results: The authors synthesized a series of novel tetrahydroquinazoline derivatives in six steps and demonstrated that their development had a unique ability to suppress HIF-1 alpha expression through proteasomal degradation. Conclusion: Among these compounds, CDMP-TQZ (8bf) exhibited the highest antiproliferative potency in human cancer cells, in part through downregulation of HIF-1 alpha.

Approximately 60% of human cancers exhibit enhanced activity of ERK1 and ERK2, reflecting their multiple roles in tumor initiation and progression. Acquired drug resistance, especially mechanisms associated with the reactivation of the MAPK (RAF/MEK/ERK) pathway represent a major challenge to current treatments of melanoma and several other cancers. Recently, targeting ERK has evolved as a potentially attractive strategy to overcome this resistance. Herein, we report the design and synthesis of novel series of fused naphthofuro[3,2-c] quinoline-6,7,12-triones 3a-f and pyrano[3,2-c]quinoline-6,7,8,13-tetraones 5a,b and 6, as potential ERK inhibitors. New inhibitors were synthesized and identified by different spectroscopic techniques and X-ray crystallography. They were evaluated for their ability to inhibit ERK1/2 in an in vitro radioactive kinase assay. 3b and 6 inhibited ERK1 with IC50s of 0.5 and 0.19 mu M, and inhibited ERK2 with IC50s of 0.6 and 0.16 mu M respectively. Kinetic mechanism studies revealed that the inhibitors are ATP-competitive inhibitors where 6 inhibited ERK2 with a K-i of 0.09 mu M. Six of the new inhibitors were tested for their in vitro anticancer activity against the NCI-60 panel of tumor cell lines. Compound 3b and 6 were the most potent against most of the human tumor cell lines tested. Moreover, 3b and 6 inhibited the proliferation of the BRAF mutant A375 melanoma cells with IC50s of 3.7 and 0.13 mu M, respectively. In addition, they suppressed anchorage-dependent colony formation. Treatment of the A375 cell line with 3b and 6 inhibited the phosphorylation of ERK substrates p-90RSK and ELK-1 and induced apoptosis in a dose dependent manner. Finally, a molecular docking study showed the potential binding mode of 3b and 6 within the ATP catalytic binding site of ERK2.

In this research, in order to synthesize a series of ethyl 2-amino-4-benzoyl-5-oxo-5,6-dihydro-4H-pyrano[3,2-c]quinoline-3-carboxylates, a green and an efficient method is proposed through one-pot three-component reaction of substituted arylglyoxals, ethyl cyanoacetate, and 4-hydroxyquinolin-2(1H)-one in the presence of terapropylammonium bromide as a catalyst in good yields. All synthesized new substances were characterized by FTIR, H-1-NMR, and C-13-NMR spectral data and elemental analysis.

Prostaglandins are among the most important natural isolates owing to their broad range of bioactivities and unique structures. However, current methods for the synthesis of prostaglandins suffer from low yields and lengthy steps. Here, we report a practicability-oriented synthetic strategy for the enantioselective and divergent synthesis of prostaglandins. In this approach, the multiply substituted five-membered rings in prostaglandins were constructed via the key enyne cycloisomerization with excellent selectivity (>20:1 d.r., 98% e.e.). The crucial chiral centre on the scaffold of the prostaglandins was installed using the asymmetric hydrogenation method (up to 98% yield and 98% e.e.). From our versatile common intermediates, a series of prostaglandins and related drugs could be produced in two steps, and fluprostenol could be prepared on a 20-gram scale.

The preparation of mesoporous poly(ionic liquid)s (MPILs) is critically fundamental for the design of heterogeneous catalysts, whereas traditional methods are difficult to obtain these materials with both well-defined mesoporous structure and unique functionality. Here, HClO4-functionalized MPILs with well-defined mesoporous structure were successfully fabricated, in which the alkene-modified Fe3O4 nanoparticles as structural reinforcer play a vital role for mesoporous structure formation. The MPILs were characterized by N-2 adsorption/desorption, scanning electron microscope (SEM), transmission Electron Microscope (TEM), Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), thermogravimetric analysis (TGA), and vibration sample magnetometry (VSM), and the results confirm that MPILs possess moderate surface area, well-defined mesoporosity, abundant active ionic centers, as well as efficient magnetic recovery. The resulting MPILs show excellent catalytic activity for condensation reaction and Knoevenagel condensation. The kinetic study reveals that the excellent catalytic activity of MPILs is attributed to the synergistic catalysis of mesoporous confinement effect and hydrogen proton from MPILs, albeit with mass transfer resistance produced by mesoporous channels. Further, the catalyst can be recovered using an external magnetic field and reused at least 10 times without a considerable decrease in its catalytic activity. Finally, alkene-modified Fe3O4 nanoparticle-mediated mesoporous construction promotes a textural engineering approach to the development of novel mesoporous materials for other applications.

Barium titanate nanoparticles (BaTiO3 NPs) were synthesized under sonochemical conditions. The synthesized BaTiO3 NPs was characterized using Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), thermogravimetry analysis (TGA), Brunauer-Emmett-Teller (BET), Field emission scanning electron microscopy (FE-SEM) and Transmission electron microscopy (TEM). Morphological study reveals that BaTiO3 NPs have spherical shape with particle size ranging between of 60 and 80 nm. BaTiO3 NPs were further utilized as an efficient and reusable heterogeneous catalyst for ultrasound assisted synthesis of pyrano[3,2-b]pyran. In addition, novel 7-tosyl-4,7-dihydropyrano[2,3-e]indole derivatives were also synthesized using ultrasonication. This new catalytic method displays several notable features such as simplicity, short reaction time, easy work up, easy purification, increased yield and green reaction conditions. The nanocatalyst was reusable up to five runs with marginal loss in its catalytic activity.

The oxidative cleavage of the C-N bond of aryl and heteroaryl (dimethylamino)methyl groups is achieved by employing molecular iodine as a mild oxidizing agent under ambient conditions in the presence of a mild base. A novel series of 7,8-dichlorobenzofuro[3,2-c]quinoline-6,9,10(5H)-triones was obtained regioselectively in good yields. The products were formed by the reactions of the 4-hydroxy-2(1H)-quinolinones with 3,4,5,6-tetrachloro-1,2-benzoquinone in tetrahydrofuran as the solvent. Due to the fact that tyrosinase is responsible for biosynthesis and regulation of melanins and browning food products, tyrosinase inhibitors can be favorable agents in cosmetics and medicinal industries. A series of novel 2-hydroxy-4-methoxybenzohydrazide were designed, synthesized, and their new application as tyrosinase inhibitors was also disclosed. Based on in vitro tyrosinase inhibitory assay, 4d as the strongest inhibitor of tyrosinase with an IC50 value of 7.57 mu M showed approximately 2.5-fold better inhibition than kojic acid as positive control followed by two compounds 4b (IC50 = 8.19 +/- 0.25 mu M) and 4j (IC50 = 8.92 +/- 0.016) which displayed preferable tyrosinase inhibitory activity. Detailed investigations on the mechanism of action of the 4d reported mix type of inhibition. More importantly, molecular modeling assessments proposed the ability of 4d for potential interaction with Cu (metal)-His (residue) within tyrosinase active site. Overall, 4d is a promising candidate for the development of anti-tyrosinase agents.

Fragile cultural relics at underwater archaeological sites are difficult to preserve in situ over long time periods due to the effects of a complex and constantly changing surrounding environment. Thus, extracting the relics from water and preserving them in a controllable environment is of primary importance. In this paper, the technology of temporary reinforcement and extraction of underwater fragile relics with veratraldehyde followed by a process of secondary temporary reinforcement using polyethylene glycol (PEG) was studied. After investigating the melting temperature and permeability of PEGs with different molecular weights, we found that PEG-3000 is an excellent candidate as the secondary reinforcing agent for the extracted relics. In the laboratory, combined with veratraldehyde and polypropylene-fiber cloth as temporary underwater strengthening materials, and selected dispersed beaded jewelry and a broken three-dimensional crock as our research subject, we carried out an entire retrieval process-temporary stabilization and secondary temporary reinforcement, to systematically evaluate the technological aspects of safely excavating fragile underwater relics in the original state. After being extracted from water and subjected to secondary temporary reinforcement using PEG-3000, the beaded jewelry and broken crock retained their underwater shape and arrangement completely. The use of secondary reinforcing agent prevents the removal process of veratraldehyde and enclosed sediment from affecting the state of the relics.
