Determination of indoles in plasma of polycystic ovarian syndrome patients by high performance liquid chromatography-fluorescence detection

A new method was established for the determination of indoles (indole (IND), 3methylindole (3 -MI), indolyl-3-acetic acid (IAA) and indolyl-3-propionic acid (IPA)) in plasma by high performance liquid chromatography-fluorescence detection (HPLC-FLD). The analytes were separated simultaneously on a Shim -pack VP-ODS column (150 mmx4. 6 mm, 4. 6 tim) using 15 mmol/L sodium dihydrogen phosphate solution and methanol (48 :52, v/v) as the mobile phases. The column temperature was 30 C, and the flow rate was 0. 8 mL/min. The calibration curves of IND, 3 -MI, IPA and IAA showed good linear relationships. The intra-day and inter-day relative standard deviations (RSDs) for the analytes were both less than 6. 31%. The average recoveries of the analytes in plasma were 97. 5% 107. 0%. The method was successfully applied to the analysis of indoles in the plasma of healthy women of reproductive age (n = 25) as controls and patients with polycystic ovarian syndrome (n = 61). The results showed that the concentrations of indoles in the plasma were significantly different between the two groups, and IND was found to be a risk factor and a potential diagnostic biomarker for polycystic ovarian syndrome (PCOS). The method is simple, sensitive and suitable for use in clinical testing and laboratory research.

Physiological impacts of zero valent iron, Fe3O4 and Fe2O3 nanoparticles in rice plants and their potential as Fe fertilizers

Fe-based nanoparticles (Fe-based NPs) have great potential as a substitute for traditional Fe-fertilizer; however, their environmental risk and impact on plant growth are not fully understood. In this study, we compared the physiological impacts of three different Fe-based NP formulations: zero-valent iron (ZVI), Fe3O4 and Fe2O3 NPs, on hydroponic rice after root exposure for 2 weeks. Fe-normal (Fe(+)) and Fe-deficiency (Fe(-)) conditions were compared. Results showed that low dose (50 mg L-1) of ZVI and Fe3O4 NPs improved the rice growth under Fe( -) condition, while Fe2O3 NPs did not improve plant growth and caused phytotoxicity at high concentration (500 mg L-1). Under Fe(+) conditions, none of the Fe-based NPs exhibited positive effects on the rice plants with plant growth actually being inhibited at 500 mg Cl evidenced by reduced root volume and leaf biomass and enhanced oxidative stress in plant. Under Fe(-) condition, low dose (50 mg L-1) of ZVI NPs and Fe3O4 NPs increased the chlorophyll content by 30.7% and 26.9%, respectively. They also alleviated plant stress demonstrated by the reduced oxidative stress and decreased concentrations of stress related phytohormones such as gibberellin and indole-3-acetic acid. Low dose of ZVI and Fe3O4 NPs treatments resulted in higher Fe accumulation in plants compared to Fe2O3 NPs treatment, by down-regulating the expression of IRT1 and YSL15. This study provides significant insights into the physiological impacts of Fe-based NPs in rice plants and their potential application in agriculture. ZVI and Fe3O4 NPs can be used as Fe-fertilizers to improve rice growth under Fe-deficient condition, which exist in many rice-growing regions of the world. However, dose should be carefully chosen as high dose (500 mg Cl in this study) of the Fe-based NPs can impair rice growth. (C) 2020 Elsevier Ltd. All rights reserved.

Unusual Formation of Cyclopenta[b]indoles from 3-Indolylmethanols and Alkynes

Acid-promoted synthesis of cyclopenta[b]indole frameworks from 3-indolylmethanols and alkynes has been reported. The overall transformation represents a formal [3 + 2] annulation via rearrangement. Characterization of zinc solubilization potential of arsenic tolerant Burkholderia spp. isolated from rice rhizospheric soil

In this study, experiments were conducted to isolate, characterize, and evaluate rice rhizosphere bacteria for their arsenic (As) tolerance ability and zinc (Zn) solubilization potential in culture media and soil. Among 20 bacterial isolates recovered, six were found to solubilize inorganic Zn salt(s) efficiently under in vitro culture conditions. 16S rRNA gene sequence-based phylogenetic analysis indicated the affiliation of efficient Zn solubilizing bacteria (ZSB) to Burkholderia vietnamiensis and Burkholderia seminalis. Zinc solubilizing efficiency (ZSE) of the bacteria varied with the concentrations and types of Zn salts used in the experiments. Increasing trend in ZSE of the bacteria was noticed when the percentage of ZnO increased from 0.1 to 0.5 but the same decreased at 1.0%. Increased Zn solubilization was noticed when bacteria were incubated with lower concentration of Zn-3(PO4)(2) and ZnCO3. In general, Zn solubilization increased with increasing incubation time in lower volume medium, while some isolates failed to solubilize one or more tested Zn salts. However, enriched concentrated cells of the ZSB in glucose amended medium with 0.5% ZnO showed an increasing trend of Zn solubilization with time and were able to solubilize more than 300 mg/L Zn. This increased rate of Zn release by the ZSB was attributed to marked decline in pH that might be due to the enhanced gluconic acid production from glucose. As evident from the decreased ZSE of the bacteria in the presence of As(V) in particular, it seems arsenic imparts a negative effect on Zn solubilization. The ZSB were also able to increase the rate of Zn release in soil. A microcosm-based soil incubation study amending the enriched bacteria and 0.5% ZnO in soil showed an elevated level of both water-soluble and available Zn compared to un-inoculated control. During Zn solubilization in microcosms, viable cells in terms of colony-forming unit (CFU) declined by the same order of magnitude both in the presence and absence of ZnO that might be due to the nutrients limiting condition aroused during the incubation period rather than Zn toxicity. The bacteria in this study also exhibited plant growth promoting traits, such as growth in nitrogen-free medium, production of indole acetic acid (IAA), and solubilization of potassium and phosphate. Our findings suggested that Burkholderia spp. could be the potential candidates for enhancing Zn dissolution in the soil that might reduce the rate of inorganic Zn fertilization in agricultural soil.
