Winter hypoxia in shallow, ice-covered lakes can be a significant limiting factor for overwintering fish populations. In this study we tested the hypothesis that low overwinter survival due to winter hypoxia is a limiting factor for a rare, adfluvial population of native Arctic Grayling Thymallus arcticus inhabiting Upper Red Rock Lake, Montana. We used a combined laboratory and telemetry study to document the extent of hypoxia over two winters and to assess the physiological tolerance, behavioral response, and winter survival in relation to hypoxia. In the laboratory, we observed a significant behavioral and physiological response to dissolved oxygen (DO) levels <= 4.0 mg/L and determined acute 24-h LC50 values (concentration lethal to 50% of test fish) of 0.75 mg/L DO for adults and 1.50-1.96 mg/L for juveniles at temperatures of 1-3 degrees C. In the field study, we observed dynamic DO concentrations (DO < 1.0 to 10.0 mg/L) during winter ice cover, ranging from persistent near-anoxic conditions near the bottom to DO concentrations > 4.0 mg/L in the epilimnion. Radiotelemetry indicated adult winter survival rate was high (0.97 in 2014, 0.95 in 2015) and that telemetered fish selected deeper (>1 m), more oxygenated habitat during ice cover. Our study demonstrated that Arctic Grayling have a high tolerance to acute hypoxia exposure and exhibit a physiological and behavioral stress response to DO concentrations <= 4.0 mg/L. Although hypoxia was present in parts of the lake, sufficient suitable habitat with DO > 4.0 mg/L was available in the lake epilimnion in both study winters. However, winter conditions more severe than those observed during our 2-year study occur periodically in the lake, and thus winterkill could still be a limiting factor for the population.

A tri-antennary Man(9)GlcNAc(2) glycan on the surface of endoplasmic reticulum (ER) glycoproteins functions as a glycoprotein secretion or degradation signal after regioselective cleavage of the terminal alpha-1,2-mannose residue of each branch. Four alpha-1,2-mannosidases-ER mannosidase I, ER degradation-enhancing alpha-mannosidase-like protein 1 (EDEM1), EDEM2, and EDEM3-are involved in the production of these signal glycans. Although selective production of signal glycans is important in determining the fate of glycoproteins, the branch-discrimination abilities of the alpha-1,2-mannosidases are not well understood. A structural feature of the Man(9)GlcNAc(2) glycan is that all terminal glycosidic linkages of the three branches are of the alpha-1,2 type, while the adjacent inner glycosidic linkages are different. In this study, we examined whether the alpha-1,2-mannosidases showed branch specificity by discriminating between different inner glycosides. Four trisaccharides with different glycosidic linkages [Man alpha 1-2Man alpha 1-2Man (natural A-branch), Man alpha 1-2Man alpha 1-3Man (natural B-branch), Man alpha 1-2Man alpha 1-6Man (natural C-branch), and Man alpha 1-2Man alpha 1-4Man (unnatural D-branch)] were synthesized and used to evaluate the hypothesis. When synthesizing these oligosaccharides, highly stereoselective glycosylation was achieved with a high yield in each case by adding a weak base or tuning the polarity of the mixed solvent. Enzymatic hydrolysis of the synthetic trisaccharides by a mouse liver ER fraction containing the target enzymes showed that the ER alpha-1,2-mannosidases had clear specificity for the trisaccharides in the order of A-branch > B-branch > C-branch approximate to D-branch. Various competitive experiments have revealed for the first time that alpha-1,2-mannosidase with inner glycoside specificity is present in the ER. Our findings suggest that exo-acting ER alpha-1,2-mannosidases can discriminate between endo-glycosidic linkages.

In spite of the enormous promise that polymeric carbon nitride (PCN) materials hold for various applications, the fabrication of high-quality, binder-free PCN films and electrodes has been a largely elusive goal to date. Here, we tackle this challenge by devising, for the first time, a water-based sol-gel approach that enables facile preparation of thin films based on poly(heptazine imide) (PHI), a polymer belonging to the PCN family. The sol-gel process capitalizes on the use of a water-soluble PHI precursor that allows formation of a non-covalent hydrogel. The hydrogel can be deposited on conductive substrates, resulting in formation of mechanically stable polymeric thin layers. The resulting photoanodes exhibit unprecedented photoelectrochemical (PEC) performance in alcohol reforming and highly selective (similar to 100 %) conversions with very high photocurrents (>0.25 mA cm(-2) under 2 sun) down to <0 V vs. RHE. This enables even effective PEC operation under zero-bias conditions and represents the very first example of a 'soft matter'-based PEC system capable of bias-free photoreforming. The robust binder-free films derived from sol-gel processing of water-soluble PCN thus constitute a new paradigm for high-performance 'soft matter' photoelectrocatalytic systems and pave the way for further applications in which high-quality PCN films are required. Due to the fact that tyrosinase is responsible for biosynthesis and regulation of melanins and browning food products, tyrosinase inhibitors can be favorable agents in cosmetics and medicinal industries. A series of novel 2-hydroxy-4-methoxybenzohydrazide were designed, synthesized, and their new application as tyrosinase inhibitors was also disclosed. Based on in vitro tyrosinase inhibitory assay, 4d as the strongest inhibitor of tyrosinase with an IC50 value of 7.57 mu M showed approximately 2.5-fold better inhibition than kojic acid as positive control followed by two compounds 4b (IC50 = 8.19 +/- 0.25 mu M) and 4j (IC50 = 8.92 +/- 0.016) which displayed preferable tyrosinase inhibitory activity. Detailed investigations on the mechanism of action of the 4d reported mix type of inhibition. More importantly, molecular modeling assessments proposed the ability of 4d for potential interaction with Cu (metal)-His (residue) within tyrosinase active site. Overall, 4d is a promising candidate for the development of anti-tyrosinase agents.

Developing efficient and selective strategies to approach complex architectures containing (multi)stereogenic centers has been a long-standing synthetic challenge in both academia and industry. Catalytic cascade reactions represent a powerful means of rapidly leveraging molecular complexity from simple feedstocks. Unfortunately, carrying out cascade Heck-type reactions involving unactivated (tertiary) alkyl halides remains an unmet challenge owing to unavoidable beta-hydride elimination. Herein, we show that a modular, practical, and general palladium-catalyzed, radical three-component coupling can indeed overcome the aforementioned limitations through an interrupted Heck/allylic substitution sequence mediated by visible light. Selective 1,4-difunctionalization of unactivated 1,3-dienes, such as butadiene, has been achieved by employing different commercially available nitrogen-, oxygen-, sulfur-, or carbon-based nucleophiles and unactivated alkyl bromides (>130 examples, mostly >95:5 E/Z, >20:1 a). Sequential C(sp(3))-C(sp(3)) and C-X (N, O, S) bonds have been constructed efficiently with a broad scope and high functional group tolerance. The flexibility and versatility of the strategy have been illustrated in a gram-scale reaction and streamlined syntheses of complex ether, sulfone, and tertiary amine products, some of which would be difficult to access via currently established methods.

Two cocoa types are predominantly being cultivated in Ecuador, the highly appreciated fine Nacional and the high-yielding and disease-resistant CCN51, classified as bulk. With the introduction of foreign germplasm in Ecuador and the international demand for high-quality Nacional cocoa, the present study aims to characterize and compare the flavor profile of Nacional hybrids (EET103, EET559, EET576 and EET577) and CCN51, and to assess the effect of the genotype. The fermented dried beans were processed into liquor and their sensory properties, volatile composition and odor fingerprint were analyzed by trained panelists, headspace-solid phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC-MS), and GC electronic nose (e-nose), respectively, combined with multivariate statistics. Seventy volatiles were identified and semi-quantified, among them, 16 and 14 volatiles conferring fruity and floral aroma notes, respectively. CCN51 was mainly characterized by undesirable odor-active volatiles, whereas the EET cultivars contained the highest concentration of most fruity and floral volatiles, with differences in between EET cultivars. Sensory data highlighted organoleptic differences between the fine and bulk liquors, i.e. the fruity and acid EET flavor strongly differed from the bitter, astringent and off-flavor attributes of CCN51. Principal component analysis (PCA) discriminated firstly between fine and bulk, and secondly between EET identity. Partial least squares analysis identified possible correlations between sensory attributes and volatile compounds. Besides, e-nose proved its usefulness for rapid assessment of the liquors’ sensory profile. The results provide a foundation for understanding the genotype’s contribution to and the compositional basis of Ecuadorian fine and bulk cocoa flavor.

The B3NO2 six-membered heterocycle (1,3-dioxa-5-aza-2,4,6-triborinane = DATB), comprising three different non-carbon period 2 elements, has been recently demonstrated to be a powerful catalyst for dehydrative condensation of carboxylic acids and amines. The tedious synthesis of DATB, however, has significantly diminished its utility as a catalyst, and thus the inherent chemical properties of the ring system have remained virtually unexplored. Here, a general and facile synthetic strategy that harnesses a pyrimidine-containing scaffold for the reliable installation of boron atoms is disclosed, giving rise to a series of Pym-DATBs from inexpensive materials in a modular fashion. The identification of a soluble Pym-DATB derivative allowed for the investigation of the dynamic nature of the B3NO2 ring system, revealing differential ring-closing and -opening behaviors depending on the medium. Readily accessible Pym-DATBs proved their utility as efficient catalysts for dehydrative amidation with broad substrate scope and functional-group tolerance, offering a general and practical catalytic alternative to reagent-driven amidation.
